主要学术成就、科技成果及创新点:

本人博士期间工作立足于毛细管纳升级液相色谱分离新方法新技术研究,致力于提高分离能力、分析重复性和可靠性,以及分离分析模式的自动化,并将这些技术应用于提高定性定量蛋白质组和蛋白质组翻译后修饰分析性能。主要包括以下几点:

(1)系统考察了进样预柱与分离柱间死体积对系统分离能力的影响;发展了一种基于磷酸基的强阳离子交换毛细管整体柱,进行在线二维色谱分离并显著提高蛋白质组鉴定覆盖率;在同一根毛细管中制备出两相毛细管整体柱并成功应用于蛋白质组样品分析;

(2)优化了反相整体柱对蛋白质组复杂样品的分离性能并提高了鉴定蛋白质数目;制作了毛细管整体柱在线电喷雾喷针,显著降低分离柱后死体积对系统分离能力的影响;制备了带反相整体柱塞的毛细管填充柱,降低毛细管填充柱小粒径填料填充难度;

(3)制作了反相-强阳离子交换两相上样预柱,进行在线全自动同位素标记、二维色谱分离和蛋白组定量分析;发展了一种“拟三重标”同位素标记蛋白质组定量的策略,成功定量了约2300个鼠脑海马区蛋白质,阐释了病理发展过程中的内在分子机理;

(4)建立了强阴离子交换上样预柱对磷酸肽“半在线”三步分级的方法和反相-强阳离子交换两相上样预柱对磷酸肽在线二维色谱分离并定量的技术;考察了在大规模磷酸肽定量中定量重复性和准确性之间的关系并建立筛选方法,对定量结果可靠性进行有效控制。

上述各项新技术新方法在本实验室和国内外合作实验室中被推广应用,在糖肽的分级鉴定,产油酵母的产油机理研究,组织工程构建软骨、肌腱蛋白表达研究,血浆多肽组学研究和肝癌组织蛋白质表达谱研究等工作中起到了重要作用;协助上海第二军医大,组织工程研究中心,和加拿大渥太华大学等合作单位完成了多项样品分析任务,相关研究成果发表在Journal of Proteome Research (IF 5.460), Molecular & Cellular Proteomics (IF 8.354), Biomaterials (IF 7.882), FEMS Yeast Research (IF 2.279)等杂志上。
