EMS诱导黄瓜离体变异及分子标记检测

【摘要】采用不同浓度的甲基磺酸乙酯(EMS)对黄瓜幼苗茎段进行处理,以茎段存活率和出芽率为指标,筛选到半致死处理组合,即4%EMS处理2 h.RAPD和SRAP分子标记分析发现,经EMS处理获得的再生苗DNA发生了变异,共检测到8个RAPD变异位点和5个SRAP变异位点.%Different concentrations of ethyl methane sulfonate (EMS) were used to treat the stem segments of cucumber seedlings in this research. Using the survival rate and bud differentiation rate of the stem segments as criteria, we obtained the half - lethal treatment combination, i.e. treating the stem segment with 4％EMS for 2 h. RAPD and SRAP analysis showed that DNA variation existed in the regenerated plantlets induced by EMS. The two markers detected eight RAPD variation loci and 5 SRAP variation loci.
