chemical shifts for aromatic carbons (d C 105.7, 138.6, 139.2,154.5), while for the second aromatic ring bearing two methoxyl groups, the corresponding signals could be as-signed (d C 112.2, 113.4, 122.2, 130.8, 149.9, 150.4). Exten-sive analysis of HSQC, 1H–1H COSY , and HMBC spectral data led to the establishment of the functional groups (Fig.2). The HMBC correlations from H-7?to C-2?and C-6?, and from H-2, H-6 to C-7, con?rmed the location of each oxy-genated aromatic ring to the tetrahydrofuran ring. The HMBC correlations from methoxyl protons to C-3, C-4, C-3?, C-4?, and C-5?indicated that the ?ve methoxyl groups are attached at C-3, C-4, C-3?, C-4?, and C-5?, respectively. Ex-cept for the presence of the above ?ve methoxyl groups, an especially up?eld signal of methoxyl group (d H 3.21, d C 50.6) was observed in 1H- and 13C-NMR spectrum, which was attached at C-7. This was established by HMBC correla-tion observed from methoxyl protons to C-7 and the relative down?eld shift of C-7 (d C 113.7, s). In addition, the HMBC correlations from H 3-9, H 3-9?, H-7?to the oxygenated quater-nary carbon C-8 (d C 83.5, s), along with the analysis of its molecular formula, indicated that C-8 of 1was substituted by a hydroxyl group. The relative con?guration of 1was shown

to be as depicted in Fig. 2b by the correlations observed in a ROESY experiment. ROESY correlations were observed be-tween H-7?and H-6?(H-2?), H 3-9 and H 3-9?; H-7?also gave a ROESY correlation to H 3-9?, which established that H-7?,H 3-9?, and H 3-9 were in b -orientation, and the ROESY corre-lations of H-8?with H-2?(H-6?), and CH 3O-7 with H-6?(H-2?) indicated H-8?and CH 3O-7 were in a -orientation. Thus the structure of 1was determined, and named as kadlongirin A. Note that compound 1is the ?rst example of a tetrahydro-furan lignan substituted at C-7 with a methoxyl group in Kadsura species.

Compound 2, obtained as yellowish oil, had the molecular formula C 24H 30O 8, as derived from HR-ES I -MS at m /z 469.1850 ([M ?Na]?, Calcd 469.1838). The UV , I R, and NMR spectra established that 2possessed tetrahydrofuran lignan skeleton,21,22)differing from 1by the loss of two hy-drogens. Detailed comparison of 1H- and 13C-NMR data of 2with those of 1showed close analogy. The major differences included the appearance of a tetrasubstituted epoxide (d C 71.3, s; 73.5, s) in 2and disappearance of a methine (d C 50.4, d) in 1. The HMBC (Fig. 3a) correlations from H 3-9and H 3-9?to C-8 and C-8?, respectively, further veri?ed that the epoxide group was positioned between C-8 and C-8?. In the ROESY (Fig. 3b) experiment, H 3-9?showed strong corre-lations with H 3-9 and H-7?, and H-7?with H-2?(H-6?), indi-cating an a -orientation of the epoxide group. CH 3O-7 (d H 3.13, s) showed correlation with H-2 and H-6?(H-2?), indi-cating its a -orientation. Thus all of these data con?rmed the structure for 2with an epoxide of a -orientation located be-tween C-8, and C-8?, named as kadlongirin B.

Compound 3was obtained as yellow amorphous powder.ts molecular formula, C 15H 20O 2, was determined by the pseudo-molecular ion peak [M ?Na]?in the positive HR-ESI-MS at m /z 255.1361 (Calcd 255.1360), indicating 6 de-grees of unsaturation. The I R spectra revealed the presence of hydroxyl group (3423cm ?1) and phenolic group (1449,1643cm ?1).23)The 1H-NMR spectrum of 1(Table 1) showed two aromatic protons (d H 6.55, s; 6.73, s), suggesting a tetra-substituted aromatic spin-system with a para con?guration.A series of complex signals at d H 4.64, 4.88 (2H, each br s),d H 3.79 (1H, ddd, 2.4, 4.8, 6.8Hz), d H 3.58 (1H, dd, 7.6,8.4Hz), and 2.65 (1H, m) was indicative of a characteristic ole?nic methylene and carbinolic and two benzyl hydrogens.Further signals at d H 1.25 (3H, d, 7.1Hz), d H 1.65 (3H, s),and 2.08 (3H, s) revealed the presence of two aliphatic groups and an aromatic methyl group in the molecule. The 13

C-NMR data (Table 2) and DEPT experiments clearly indi-cated the basic C 15-sesquiterpene skeleton of the cadinane skeleton.24,25)The structure of 3was elucidated from HSQC,1

H–1H COSY , and HMBC studies (Fig. 4a). The HMBC cor-relations of H 3-15 to C-2, C-3, and C-4, and H-4 to C-2, con-
