大豆贮藏蛋白序列分析与3-D结构同源模建

大豆贮藏蛋白序列分析与3-D 结构同源模建

摘要:分析确定大豆贮藏蛋白的种类二序列同源性二进化关系二理化特征和空间结构三以大豆(Glycine max Wm82.a2.v1)基因组为材料,采用数据库在线工具和生物信息学软件对大豆贮藏蛋白基因的染色体定位,蛋白序列信息进行分析预测,并对其3-D 结构进行同源模建三大豆贮藏蛋白基因具有多拷贝和单基因多种转录拼接方式并存的遗传模式,其蛋白序列相似度较高,系统进化树分析表明其聚为5支,对应5种类型蛋白质三除7S 碱性球蛋白外,其他贮藏蛋白的理化性质接近,含有较高比例的鲜味氨基酸三大豆贮藏蛋白空间结构各异,11S 球蛋白二7S β-伴球蛋白和碱性球蛋白分别以六聚体二三聚体和四聚体形式存在,2S 清蛋白表空间结构松散,由α-螺旋和无规卷曲构成三该研究可为大豆贮藏蛋白的深入研究以及大豆加工食品二大豆酿造调味品的开发提供理论参考三

Abstract :To analyze and confirm the species,sequence homology,evolutionary relationships,physico-chemical pa-rameters and 3-D structures of soybean storage proteins.Glycine max Wm82.a2.v1genome was served as matrial,online tools of database and bioinformatics softwares were applied to analyze or predict the chromosome localization of genes and sequence information of soybean storage proteins.Genetic model of multi-copy genes and multi-tran-scription splicing for a gene both existed in soybean storage proteins,which had a high sequence homology.Phylo-genetic tree analysis showed that soybean storage proteins belonged to 5clusters,corresponding to 5kinds of pro-teins.The soybean storage proteins,except basic 7S glycinin,with a similar physico-chemical parametaers contained a higher proportion of umami amino acid.The 3-D structure of soybean storage proteins were various,11S glycinin,7S β-conglycinin and basic 7S glycinin corresponded to hexamer,trimer,tetramer,respectively.3-D structure of

2S albumin loose constituted with α-helix and random coil.This study could provide therotical basis for deep re-search of soybean storage proteins and for development of soybean processed foods and fermented flavouring foods.Key words :soybean;storage proteins;sequence analysis;homology modeling
